Beschrijving van insuline Lispro, recombinant
Insuline lispro is een snelwerkende insuline die een analoog van humane insuline bevat met de aangepaste aminozuursequentie aan de B-keten, met de chemische naam Lys(B28), Pro(B29) humane insuline-analoog.
Formuleringen op basis van insuline lispro zijn sinds 1996 op de markt verkrijgbaar onder de merknaam Humalog (Lilly); en andere beschikbare producten zijn Lyumjev (Lilly) en Admelog (Sanofi).

Productieproces van insuline Lispro
Stroomopwaarts proces
- E. coli-stam expandeerde in een schudfles uit een flesje Working Cell Bank (WCB);
- Fermentatie: zaadfermentatie; en inductie van de expressie van insuline lispro door fermentatie met hoge dichtheid.
Stroomafwaarts proces
- Het oogsten van E. coli-cellen en inclusielichamen door centrifugatie en cellyse.
- Hervouwen van inclusielichaampjes om de voorloper van insuline lispro te produceren;
- Enzymatische splitsing en zuivering om zuivere werkzame stof te verkrijgen.
Vullen en voltooien
- Voorbereiding van de formulering en aseptische afvulling in injectieflacons of voorgevulde cartridges.
