Interferon-α (IFN-α)

Modality

Interferons (INFs) are cytokines secreted during the early stages of viral infection that have antiviral and antiproliferative effects. Three types of INFs are present in the human body: IFN-α, IFN-β, and IFN-γ. The IFN-α subtypes is the most widely used INF for therapeutic purposes.

Recombinant IFN-α and PEG-conjugated IFN-α are currently approved for curing patients suffered from viral infections and cancers, such as hepatitis B, hepatitis C, and malignant melanoma.

Recombinant IFN-α is typically produced in Escherichia coli (E. coli) and is currently used in clinical practice because it does not require post-translational modification to exhibit biological activity.

Application of Interferon-α (IFN-α)

Interferon alfa-2a (IFN-α-2a)

Interferon alpha-2a, the active substance of Roferon-A, is a highly purified protein with a molecular weight of approximately 19 kDa and contains 165 amino acids. Developed by Roche and manufactured using recombinant DNA technology, roferon-A applies a genetically engineered E. coli containing DNA that codes for the human protein.

Peginterferon alfa-2a

Pegasys (peginterferon alfa-2a) is a covalent linker consisting of recombinant interferon alfa-2a (approximately 20 kDa) and branched bis-monomethoxy polyethylene glycol (PEG) chains (approximately 40 kDa). Through a stable amide bond to lysine, the PEG moiety binds to the interferon alfa moiety at a single site. Interferon-alpha-2a is manufactured by recombinant DNA technology in which the cloned human leukocyte interferon gene is inserted into and expressed in E. coli. The Pegasys formulation was developed by Roche/Genentech and acquired by zr pharma worldwide (except China and Japan).

Interferon alfa-2b

Produced by recombinant DNA technology, Intron-A (interferon-alpha-2b) is a purified, sterile recombinant alpha interferon product, which is produced by bacterial fermentation of E. coli strain carrying a genetically engineered plasmid that contains the interferon alfa-2b gene extracted from human leukocytes. And intron A was developed by Schering-Plough and Merck Sharp & Dohme.

Peginterferon alfa-2b

Pegintron (peginterferon α-2b) is a covalent bond consisting of recombinant interferon α-2b and monomethoxy polyethylene glycol (PEG). With a molecular weight of 19 kDa, Interferon α-2b is a water-soluble protein and developed by recombinant DNA techniques. Developed by Schering-Plough and Merck Sharp & Dohme, it is produced by bacterial fermentation of an E. coli strain carrying a recombinant plasmid containing the human leukocyte interferon gene.
