mRNA Vaccines for Cancer Therapy

Currently, cancer vaccines have been given great expectation to be a possible therapy for cancer treatment due to their specificity, safety, and tolerability. mRNA cancer vaccine contains three critical components: antigen and adjuvant for immune stimulation, and drug delivery system. mRNA cancer vaccines deliver mRNA encoding anticancer antigens to host cells (usually antigen-presenting cells (APC)), causing them to express the target protein antigens, and then the major histocompatibility complex (MHC) is presented on the surface of the APC, resulting in the desired immune response.

There are some mRNA-based vaccines encoding Tumor Associated Antigens (TAA) and Tumor Specific Antigens (TSA) are in clinical trials in patients with lung cancer, colorectal cancer, melanoma cancer, head and neck cancer, renal cancer, liver cancer, glioma, or solid cancer.
