Description of Insulin Glargine, Recombinant

Insulin glargine is the first long-acting basal insulin (once daily injection). It is a human insulin variant with three site differences: one amino acid substitution Gly(A21), and two arginines (Arg) added at the C-terminal of the B chain.

Insulin glargine was developed by Sanofi and approved in 2000 for diabetes mellitus. Recently, available insulin glargines include Lantus (Sanoﬁ), Toujeo (Sanoﬁ), Basaglar (Lilly), Rezvoglar (Lilly), and Semglee (Mylan).

Amino Acid Sequence of Insulin Glargine

Insulin glargine differs from native human insulin at one amino acid of the A chain [Gly(A21)], and two amino acids at the C-termini of the B chain (Arg-Arg added).
