MEPE Fragment

Matrix extracellular phosphoglycoprotein (MEPE) is part of the family of short integrin-binding ligand interacting glycoprotein (SIBLINGs), which perform important roles in the biomineralization process. And the main source of MEPE is bone cells (osteoblasts, osteocytes, and odontoblasts).

MEPEs contain a conserved Arg-Gly-Asp (RGD) motif, which is thought to be involved in integrin recognition. AC-100 (Dentonin) is reported to be the mid-terminal part of MEPEs and contains RGD and Ser-Gly-Asp-Gly (SGDG) sequences stimulating new bone formation in vitro and in vivo. RGD and SGDG have been identified as integrin-binding and glycosaminoglycan-attachment motif, respectively.

AC-100 (Dentonin), a novel 23 amino acid synthetic peptide originating from a fragment of human MEPE, has been provided to promote bone formation and osteoblast proliferation. AC-100 was developed by Acologix, Inc, whose amino acid sequence is TDLQERGDNDISPFSGDGQPFKD. Recombinant MEPE fragments or MEPE fragments fusion protein produced by recombinant DNA technology is also being studied.
