mRNA is synthesized by in vitro transcription (IVT) from a linearized plasmid DNA (pDNA). Some by-products are also formed during IVT procedure, including double-stranded RNA (dsRNA) identified as major triggers for the innate immune pathway. It is critical to remove impurities and obtain high yields of pure mRNA for in vitro/vivo mRNA research and therapeutics applications.
Yaohai Bio-Pharma has established a set of mRNA synthesis technologies to provide highly pure mRNA, such as sequence design and optimization, IVT, purification, lyophilization and lipid nanoparticle (LNP) encapsulation. All products are released under the stringent quality control standards. Affinity chromatography (AC), based on the strong and selective interaction between 3′ polyA tail of mRNA with stationary phase oligo dT, is an efficient and scalable purification strategy for mRNA.
