Firefly luciferase (F-Luc) mRNA expresses a luciferase protein, originally isolated from the firefly, Photinus pyralis. This protein emits bioluminescence in the presence of the substrate, luciferin.
Firefly luciferase mRNA is commonly used as positive control in mRNA transfection and delivery system development. Firefly luciferase can also be utilized in mammalian cell culture to measure both gene expression and cell viability.

Taking linearized plasmid DNA (pDNA) harboring F-Luc mRNA coding genes as IVT template, we obtained pure F-Luc mRNA transcripts by co-transcriptional capping and purification, with cap1 structure and N1Ψ modification.
Pure F-Luc mRNA (1 μg) mixed with a commercial transfection reagent was transfected into 293T cells in a 96 well plate. And luciferase assay was performed after 24 hours.
