eGFP mRNA expresses enhanced green fluorescent protein (eGFP), originally derived from the jellyfish, Aequorea victoria. eGFP displays bright green fluorescence when exposed to light in the blue to ultraviolet range, with an emissions peak at 509 nm. eGFP mRNA is commonly used as reporter marker (positive control) in transfection and delivery system development.

Taking linearized plasmid DNA (pDNA) harboring eGFP mRNA coding genes as IVT template, we obtained pure eGFP mRNA transcripts by co-transcriptional capping and purification, with cap1 structure and N1Ψ modification.
Pure eGFP mRNA (1 μg) mixed with a commercial transfection reagent was transfected into 293T cells in a 96 well plate. And fluorescence photography was performed after 24 hours.
