Explore the Fascinating World of the Lithium Chloride Precipitation

Lithium chloride precipitation is widely used for separating RNA from solution.

Here's the breakdown of the process:

1. Sample preparation: Start with a sample containing RNA, such as in-vitro transcript mixture.

2. Lithium chloride addition: Add an appropriate volume of concentrated lithium chloride solution to the RNA solution.

3. Incubation: After adding lithium chloride, the solution is incubated at a low temperature, generally around -20°C. This causes RNA molecules to precipitate out of the solution.

4. Centrifugation: After incubation, the solution is centrifuged at high speed. This separates the precipitated RNA from the rest of the solution, which contains impurities.

5. Washing: Once centrifugation is complete, carefully remove the supernatant but without interfering with the RNA precipitation. Wash the RNA particles with cold ethanol or isopropyl alcohol to remove any remaining impurities.

6. RNA resuspension: After washing, the RNA precipitate is briefly air-dried and then re-suspended in an appropriate buffer, such as TE buffer (Tris-EDTA buffer) or distilled water. Gentle pipetting or eddy currents may be necessary to adequately resuspend the RNA.

7. Quantification and analysis: Finally, the isolated RNA is quantified by ultraviolet spectroscopy or fluorescence method. This determines the concentration and purity of the RNA sample, which is important for downstream applications such as LNP encapsulation, and cell transfection.
